Lumen cues calcium release

چکیده
Lumen cues calcium release alcium release from lumenal stores is sensitive to the redox state of the ER lumen, as shown by Takayasu Higo, Katsuhiko Mikoshiba (University of Tokyo, Japan), and colleagues. The group has identified the first ER lumenal protein that controls IP 3 -regulated calcium channels (IP 3 Rs). The opening of IP 3 Rs is tightly controlled by cytosolic conditions. Most scientists assume that conditions in the ER also regulate channel opening, but the evidence was minimal. To find any relevant regulators in the ER, the authors used a lumenal domain of IP 3 R1 to fish out binding proteins. They discovered ERp44, whose association with IP 3 R1 blocked IP 3 -induced calcium release. But the binding of ERp44 and IP 3 R1, and subsequent channel blockage, depended on several conditions. First, the association occurred at low calcium levels, as would be seen after ER calcium stores are depleted. Second, it required that the lumenal domain of the channel be in its reduced state, with free thiol groups. That ERp44 is sensitive to the redox state is not surprising— C

Calcium Sparks in Cardiac Cells in Silico
We simulate elementary calcium release events (sparks) in a single calcium release unit in ventricular myocyte. Previously developed and tested electron-conformational model of the stochastic dynamics of RyR-channels is integrated to the calcium dynamics model in the cardiac cell. This approach allows to observe RyRs opening/closing in details on the macromolecular level during the calcium dyna...
متن کاملTriadin, a linker for calsequestrin and the ryanodine receptor.
Protein components of the triad junction play essential roles in muscle excitationcontraction coupling (EC coupling). Considerable research has been performed on the identification and characterization of proteins that regulate calcium storage and release from the sarcoplasmic reticulum (McPherson and Campbell, 1993; FranziniArmstrong and Jorgensen, 1994). Key proteins characterized include the...
متن کاملCalcium release through P2X4 activates calmodulin to promote endolysosomal membrane fusion
Intra-endolysosomal Ca(2+) release is required for endolysosomal membrane fusion with intracellular organelles. However, the molecular mechanisms for intra-endolysosomal Ca(2+) release and the downstream Ca(2+) targets involved in the fusion remain elusive. Previously, we demonstrated that endolysosomal P2X4 forms channels activated by luminal adenosine triphosphate in a pH-dependent manner. In...
متن کاملCharacterization of transepithelial potential oscillations in the Drosophila Malpighian tubule.
The Malpighian tubule of Drosophila melanogaster is a useful model system for studying the regulation of epithelial ion transport. In acutely isolated tubules, the transepithelial potential (TEP) undergoes large oscillations in amplitude with a period of approximately 30s. The TEP oscillations are diminished by reductions in the peritubular chloride concentration in a manner consistent with the...
متن کاملExcitation-contraction coupling from the 1950s into the new millennium.
1. Excitation-contraction coupling is broadly defined as the process linking the action potential to contraction in striated muscle or, more narrowly, as the process coupling surface membrane depolarization to Ca(2+) release from the sarcoplasmic reticulum. 2. We now know that excitation-contraction coupling depends on a macromolecular protein complex or 'calcium release unit'. The complex exte...
